Photoshop has so many ways to get the job done its insane but any decent raster software can do the same. I use to hate the magic wand but I've seen its powers when used appropriately.

the bucket method i would recommend for smaller images, but for real complex shapes on large images the pen tool can get clean scalable results.

It was cool because it allowed filters and layers with indexed color and it had some nice effects and options for a cheap app that came with my old SCSI scanner. I want to get into photoline, which is actually a lot more robust than photoshop in some regards... but that interface...

So I downloaded a Netflix rip of the episode (no seeds for the DVDrips) and as kids they're never in natural light. But at the end of the episode they are and I get a skin tone of #856E5F. I only downloaded the episode "Kid Stuff" so I couldn't look for any other examples of Wonder Woman in natural light right now. But maybe someone else wants to go ahead and download a few episodes and look. I did however do a Google image search for a character sheet and found this http://tinyurl.com/oz3f3ku which honestly shows the colors I remember but it doesn't look like an official picture so the colors there are not 100% reliable. On there I get #F9D5C1. So use one of those or do your own research.

Gimp has a 'color to alpha' option that I think does this in one step, is there a Photoshop equivalent?
I'm currently using it to add coloured lines to that Gwen v Wonderloli: Dawn of Cuteness series; is indeed very useful.

Yeah, extracting the lines is easy. just go to channels and hit the button to load the channel as a selection. Everything that's white (and some light greys) will be selected while blacks (and darker greys) will be preserved.

One thing that I like to do is after my flats and shades are done, I like to combine the flats layers into one layer, bring move that layer to the top of my linework layer, set the colors layer to hard light and use Image>adjustments>hugh/saturation to darken it until the colors appear to be the same as they were when they were under the linework. This helps add a little hint of color to the linework itself based on which colors overlap which lines. Another way is to simply highlight the linework itself and paint over the lines with the desired colors on a new layer above the artwork (like in your linked layer example). That's a technique that I taught to an associate of mine a few years ago when he was first getting into photoshop and his colors are incredible these days.

i mapped the enter key to my mouse right scroll button after figured out that im a hand on stick and throttle player.

it helps with dealing with photoshop/windows operations that use popup windows or dialogue boxes, i don't need to aim to click ok; i just make the adjustment and nudge on the scroll wheel.
